- W1994936984 abstract "Activity-dependent neuroprotective protein (ADNP) is vital for brain formation and neurite outgrowth. Davunetide (NAPVSIPQ), a neuroprotective peptide derived from ADNP, protects against ADNP deficiencies (1, 2). In vivo, davunetide improves cognitive functions (e.g. spatial visual memory) and reduces tau pathology in relevant models of Alzheimer's disease and related tauopathies (3, 4), suggesting an effect on the microtubule system. The neuronal differentiation model, rat pheochromocytoma (PC-12) cells were subjected to in vitro protein immuno-quantitation, as well as quantitative real time polymerase chain reaction to measure effects of davunetide treatment on the microtubule system. Davunetide treatment significantly affected beta III tubulin expression, and the alpha tubulin tyrosination cycle in PC-12 cells. With tubulin tyrosination being important for axonal-dendritic specificity (5) and beta III tubulin being a subject for deleterious post-translational modifications in Alzheimer's disease (6), davunetide effects on the tubulin pool in cells implicate neuronal plasticity and neuroprotection. In phase II clinical trials, davunetide improved memory scores in subjects with amnestic mild cognitive impairment (7) and is currently in clinical development for frontotemporal dementia (www.allontherapeutics.com).
