- W2596709327 abstract "Rheumatoid arthritis is a systemic, inflammatory autoimmunedisorder that is manifested as a symmetric polyarthritisassociated with swelling and pain in multiple joints.Generally, activated macrophages are involved inpathophysiology of inflammation. The aim of our present studywas to compare a distribution of ED1+ activated macrophages inthe lumbar dorsal root ganglia and changes of their amount intime. We used a rat model of unilateral antigen-inducedarthritis (AIA). For induction of AIA, rats were immunized byan intra-articular injection of antigen methylated bovine serumalbumin in saline emulsified with CFA into the knee joint. Theknee joints of control rats were injected by saline and thenaive rats were without AIA induction and injection of saline.All animals were left to survive for 4, 7, 14 and 21 days.Invasion of ED1+ macrophages was detected and quantifiedbilaterally in the lumbar dorsal root ganglia (L3-L5) usingumage analysis system (NIS Elements). The data differences wereevaluated by Mann-Whitney test using Statistica software. ED1+macrophages were found bilaterally in lumbar L3-L5 dorsal rootganglia, especially in the L3 and L4. Four days after inductionof AIA, we observed robust increasing of ED1+ macrophages inthe ipsilateral lumbar ganglia. In contrast, contralateralganglia have shown only a slight increase in the number of ED1+macrophages. Seven and fourteen after immunization, allinvestigated dorsal root ganglia were infiltrated byapproximately the same numbers of ED1+ macrophages. Twenty onedays after AIA, i.e. during the chronic phase, we found noincreased infiltration of ED1+ macrophages in ipsilateraldorsal root ganglia when compared to control and naive rats.However, in contralateral dorsal root ganglia we foundapproximately the same number of macrophages like seven orfourteen days after immunization. In summary, we observedbilateral invasion of ED1+ macrophages into dorsal root gangliaaccompanying the acute, but not the chronic stage of AIA. Thepresent study has shown that acute AIA is associated with abilateral invasion of activated ED1+ macrophages into lumbardorsal root ganglia involved in afferent innervation of theknee joint." @default.
