Asymmetric catalytic [2+2+2] cycloaddition reaction involving nitriles enables straightforward access to complex chiral pyridines. This perspective highlights the recent advances in transition-metal-catalyzed asymmetric [2+2+2] cycloaddition of two alkynes and one nitrile to construct pyridine rings. Specifically, the discussion covers (1) assembling axial-chiral aryl-substituted pyridines by cobalt or rhodium catalysis and (2) manufacturing central-chiral (mainly all carbon quaternary center-containing) pyridines by rhodium-, nickel-, or cobalt-catalyzed reactions. The limitations of the current methods and the future of the field are discussed in the final section. Challenges and opportunities: • Optically active and highly substituted pyridines are of wide applications in chemistry-related fields, while their synthetic approaches are underdeveloped. • Transition-metal-catalyzed asymmetric [2+2+2] cycloaddition of one nitrile and two alkyne moieties enables straightforward and step-economical access to densely functionalized pyridines. • Despite a handful of breakthroughs being achieved, further evolution of new catalytic systems should address the efficiency regarding the reactivity and regio- and enantioselectivity, especially for fully intermolecular reactions, in order to facilitate the modular synthesis of functional group-enriched pyridine architectures. This perspective highlights the power of making enantioenriched complex pyridine-based molecules by transition-metal-catalyzed asymmetric [2+2+2] cycloaddition of two alkyne moieties and one nitrile.
