- W938295995 abstract "Thioacetylating reagents—methyldithioacetate—as sequencing reagents are reported in this chapter as illustrative examples in sequential degradation on solid-phase supports. In sequential analysis by thioacetylating reagents, peptides linked to a solid support through their COOH terminus are converted into their N-thioacetyl derivatives using dithioacetic acid esters as reagents. The thioacetyl peptides are subsequently split off in trifluoroacetic acid (TFA) yielding the 2-methylthiazol-5(4H)-one, which identifies the NH2-terminal amino acid and the resin-bound shortened peptide. The sequencing operations differ from those of the solid-phase Edman degradation in that all the reactions take place entirely within organic solvents. Dimethylformamide is chosen for the condensation step to swell the polystyrene solid support and triethylammonium acetate to speed up the thioacetylation reaction. Because thioamides are susceptible to oxidation, the maintenance of reductive conditions is important until the cyclization step; these conditions are assured during the condensation step by liberation of methylmercaptan that parallels the aminolysis of the reagent, while some mercaptoethanol is systematically added to the washing solvents." @default.
