• W2078552912 abstract "The large otolith and ocellus pigment cells of the larvae of two ascidian species, Styela partita and Ciona intestinalis, were used to investigate the chemical nature of the precursor and product of melanin synthesis and the role of these molecules in melanin granulogenesis. Phenylthiourea, an inhibitor of the enzyme tyrosinase, prevented melanin synthesis in the ascidian pigment cells. Inhibitor reversibility experiments, histochemical tests, and radioautographic examination of C14-tyrosine incorporation demonstrated that both tyrosinase and a tyrosine-rich precursor material became concentrated and localized in a granule-like vacuole of the otolith pigment cell of embryos reared in phenylthiourea. Histochemical tests, C14-labeled amino acid incorporation experiments, and inhibition of protein synthesis with puromycin and chloramphenicol have shown that the melanin in ascidian pigment cell granules is not synthesized as a melanoprotein and that the precursor material is not a tyrosine peptide. Free tyrosine appears to be the precursor which is stored in the vacuole of phenylthiourea-inhibited otolith pigment cells. This investigation defines two functionally distinct processes as part of melanin granulogenesis: appearance and activity of the tyrosinase enzyme, and accumulation of precursor tyrosine.
