- W3197014719 abstract "1898 Objectives: Combining the high spatial and temporal resolution of Magnetic Resonance Imaging (MRI) with the outstanding sensitivity of Positron Emission Tomography (PET) provides a powerful tool for molecular and cellular imaging. Therefor we synthesized a multimodal iron oxide particle and tested its detectability in vivo. Methods: An aminosilan-coated nanoparticle (MagForce®) was covalently linked with the positron-emitting radionuclide Gallium-68. These particles were injected intravenously into healthy Wistar rats (n=5). PET-imaging was performed directly p.i. using a small-bore animal-PET. A dynamic series (6 x 5 min.) was performed combined with an emission study of 30 minutes. The same animal underwent 1.5 h and 24 h p.i. MRI at 3.0 T applying a T2 weighted fast recovered fast spin echo sequence (frFSE). Results: In MRI a slight hypointensity of the liver after 1h and a significant signal drop after 24h could be detected. In animal-PET distinct radionuclide distribution in the upper abdomen was detectable after 10 minutes. Activity increased during the next 50 minutes. A definite anatomic-correlation could be verified by digital image fusion demonstrating the stability of covalent bondage under in vivo-conditions. Conclusions: This study shows the feasibility of covalently linking Aminosilan-coated nanoparticles with positron-emitting radionuclides. Due to its multiple free ammonium bonds further modifications seem to be possible.
