- W4310060200 abstract "In vitro propagation is an effective alternative technique for obtaining plant metabolites of medicinal importance. The objective of the present study was to evaluate the phytochemical accumulation, antioxidant activity and colchicine content of different organs (leaf, stem and tuber) and their correlation of both in vivo and in vitro grown Gloriosa superba L. MS media supplemented with different concentrations and combinations of BAP (1.0-3.0 mg/L) + NAA (0.2 mg/L) and Kin (1.0-3.0 mg/L) + IBA (0.2 mg/L) were used for rapid in vitro shoot multiplication and tuberization. Total phenolic and flavonoid content were estimated and the antioxidant activity was assessed using 2,2′-diphenyl-1-picrylhydrazyl (DPPH), 2,2´-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power (FRAP) and total antioxidant activity. Further, the colchicine was identified and quantified using RP-HPLC. Results revealed that maximum shoot multiplication and tuberization were obtained on MS medium supplemented with 2.0 mg/L BAP + 0.2 mg/L NAA followed by subculture with 2.0 mg/L Kin + 0.2 mg/L IBA. The maximum total phenolic, flavonoid and antioxidant activity were recorded from in vivo grown leaf. The highest amount of colchicine was obtained from in vitro grown tuber extract (2.71 ± 0.09 mg/g DE). A significant correlation was found among antioxidant activities, total phenolic and flavonoid content in case of both in vivo and in vitro grown plants. Total phenolic and flavonoid content were negatively correlated with the IC50 (DPPH and ABTS) values. However, total phenolic and flavonoid content were positively correlated with the FRAP and total antioxidant activity. Furthermore, negative correlation was obtained between total phenolic, flavonoid and colchicine content. The results suggest that in vitro grown G. superba could be a sustainable source of high antioxidant and colchicine content." @default.
