Intracellular receptors by virtue of being ligand-regulated transcription factors have naturally been examined as regulatory tools. The application exploits the normal regulatory activity of the hormone-binding domain (HBD) of vertebrate steroid receptors for the regulation of heterologous proteins in cis. On fusion to a heterologous protein, the HBD can function as an autonomous regulatory cassette and subject various protein functions to hormonal control. Thus, one or several functional activities of the heterologous moiety may be turned off in the absence of steroid ligand. While the chimeric protein can be expressed constitutively, it becomes active only on addition of hormone. Other methods to obtain conditional expression and/or activity of proteins of interest are also described in the chapter." @default.
