- W1003814080 abstract "This chapter discusses the chromatography of proteins and peptides on diethylaminoethyl (DEAE) cellulose. DEAE-cellulose is an anion exchanger with residues linked to the hydroxyl groups of the cellulose. DEAE-cellulose is particularly useful for acidic polyelectrolytes and offers the advantages of high capacity, good resolving power, and a minimum of surface effects which tend to denature proteins. Elution of the protein from DEAE-cellulose can be achieved in three ways: (1) reduction in the pH of the eluent, which lowers the net negative charge on the protein, thus weakening the binding to the exchanger; (2) increasing the pH of the eluent, thus suppressing the ionization of the basic groups of the absorbent and reducing its anion-binding capacity; and (3) increasing the ionic strength of the eluent, which decreases the strength of the ionic interactions between the protein and the exchanger. The backbone of DEAE-cellulose is made up of polysaccharide chains held together by hydrogen bonds. DEAE-cellulose will bind carbonate and bicarbonate anions that are introduced by prolonged exposure of a suspension of the resin at alkaline pH." @default.
