- W1004080392 abstract "Publisher Summary This chapter describes the properties of two new ouabain derivatives that can be used to covalently label the digitalis binding site. One of them, p -nitrophenyltriazene ouabain (NPT-ouabain), is an alkylating agent. The alkylation reaction involves a proton transfer from the receptor protein. The second molecule, aryldiazonium ouabain (AD-ouabain), is a photoactivable reagent. It differs from other photoactivable derivatives of ouabain or other digitalis-like compounds by the fact that it is possible with that derivative to photoactivate selectively only the molecules of AD-ouabain that are specifically bound to the digitalis site of the Na, K-ATPase molecule. This activation involves energy transfer through the excitation of tryptophan residues of the receptor protein. Under the conditions provided in the chapter, free AD-ouabain molecules are not photoactivated. The main properties of the covalent labeling of the digitalis receptor by NPT-ouabain and AD-ouabain are the following: (1) the percentage of covalent incorporation is higher than 20%, (2) [ 3 H]NPT-ouabain and [ 3 H]AD-ouabain specifically label the α-subunit of purified Na, K-ATPase from rabbit kidney and from the electric organ of Electrophorus electricus ." @default.
