• W100603968 abstract "IL-6 secretion was investigated in human cultured lung fibroblasts, stimulated with IL-1β. IL-1β stimulated the IL-6 secretion in a dose- and time-dependent manner. The transcriptional factor NFκB has been claimed to be involved in the expression of the IL-6 gene after IL-1 stimulation and its activation could be mediated, at least partially, by free radicals produced as second messengers in response to the IL-1 signal. So, we first checked if the NFκB factor was indeed activated in our model of human lung fibroblasts after IL-1 stimulation, using the electrophoretic mobility shift analysis (EMSA). As this was indeed the case, we further investigated whether oxidative mechanisms could be involved in the activation of NFκB. A indirect approach was chosen: cells were stimulated with IL-1 in the presence of various antioxidant molecules and their effect tested on the secretion of IL-6. Two thiol-reacting molecules, pynolidine dithiocarbamate (PDTC) and N-acetyl-cystein (NAC) were able to completely inhibit the IL-6 secretion in a dose-dependent manner. Trolox C, an hydrosoluble analogue of vitamin E also completely inhibited the IL-6 secretion, while desferrioxamine had no effect at least at the concentration used. These data strongly support the involvement of oxidative mechanisms in the activation of NFκB leading to IL-6 secretion, in human lung fibroblasts stimulated with IL-1." @default.
