- W1006298314 abstract "This chapter discusses the immobilization of steroid-transforming microorganisms in polyacrylamide. Because acrylamide polymerization is an exothermic reaction, means must be provided to dissipate the developed heat that might otherwise denature the steroid-converting enzyme system. A sandwich such as polymerization vessel having good heat-transfer properties was prepared from two thin-layer chromatography glass plates and a piece of latex tubing. The glass plates were kept 1–2 mm apart by the latex tubing and were firmly pressed against the tubing by means of metal clamps. The volume of the vessel could conveniently be adjusted between 10 and 60 ml by merely repositioning the latex tubing. The construction of the vessel also allowed easy transfer to cooling-baths of desired temperatures. The gel sheet is removed from the polymerization vessel and granulated either by passing it through 30-mesh nylon net or by treating it in a homogenizer together with buffer. Both methods produce fines, which may be removed by decantation. The gel granules are poured into a column and washed overnight with buffer to remove any remaining monomers and cell debris." @default.
