- W1010489865 abstract "建立了高效液相色谱-质谱法测定细胞样品中多巴胺（DA）、1-甲基-6,7-二羟基-1,2,3,4-四氢异喹啉（Sal）和N-甲基-6,7-二羟基-1,2,3,4-四氢异喹啉（NMSal）含量的检测方法。细胞样品经超声破碎、离心取上清、1mol/L高氯酸溶液除蛋白并再次离心取上清后,用高效液相色谱-质谱测定,选择母子离子对多离子反应监测（MRM）,内标法定量。采用DiscoveryHS F5柱（150mm×2.1mm,3μm）;流动相为5mmol/L甲酸铵缓冲溶液（pH=3.0）-甲醇（80∶20,体积比）。DA、Sal和NMSal在0.5～100μg/L范围内线性关系良好（r〉0.9995）,检出限分别为0.20、0.34、0.33μg/L,日内和日间精密度（以相对标准偏差表示,RSD）均低于8%,回收率分别为91.4%～109.6%、87.4%～109.7%、88.0%～102.7%。用该方法检测经H2O2诱导的神经母细胞瘤细胞SH-SY5Y,发现DA和Sal的含量与对照组相比都有显著增加。方法选择性好,灵敏度高,适用于测定细胞样品中DA、Sal和NMSal的含量的相关研究。" @default.
