- W101164856 abstract "Changes in O-linked attachment of N-acetyl-glucosamine (O-GlcNAc) on nucleocytoplasmic protein plays an important role in the regulation of cell function and is associated with the development of disease. Although we have recently described changes in O-GlcNAc in the rat heart from adolescence to adulthood, there is very little known about the effect of aging on O-GlcNAc levels in the heart. Therefore the goal of this study was to determine whether the development of senescence is associated with changes in the regulation of O-GlcNAc in the cardiovascular system. Tissues were harvested from 5 and 24 month old male Brown-Norway rats and O-GlcNAc levels were increased with age in the heart (207±33%) and aorta (164±18%) compared to the younger animals; surprisingly this was associated with decreased O-GlcNAc transferase (OGT) expression in heart (69±4%), and aorta (71±5%). At 24 months the increase in O-GlcNAc in hearts was associated with increased glutamine:fructose-6-phosphate amidotransferase-2 mRNA (480±100%) and UDP-N-acetylglucosamine levels (50.8±1.7 vs. 67.4±6.8 nmol/g tissue wet weight). There were similar age-dependent changes in O-GlcNAc and OGT in skeletal muscle and brain but not liver. Increased O-GlcNAc levels have been associated with impaired cardiomyocyte function in diabetes; thus this increase in O-GlcNAc seen here may contribute to the increase in cardiovascular dysfunction seen with age." @default.
