- W1013048253 abstract "Successive pathologic observations on main tissues and organs of cultured scallop Chlamys farreri, collected from the coast of Shandong (China) during mass mortality due to the onslaught of virulent disease, were conducted with light microscope and transmission electron microscope in 2000~2002. The observation of the moribund scallops showed that distinct histopathological changes appeared. The lesions were found in gills, mantle, kidney, intestine and digestive gland, but adductor muscle and sexual gland were excepted. The typical infection was characterized as multiple and severe infection with disordered and necrotic architecture. Indistinct basophil inclusions were observed on the infected tissues. Cellular changes included nuclear hypertrophy, chromatin margination, pyknosis, karyorrhexis, and cellular disintegration that often resulted in empty space amongst the lesions. Transmission electron microscope revealed the presence of sphere-shaped, enveloped virions, 130 to 170 nm in diameter, in the cytoplasm of infected cell from above-mentioned various tissues. The spike of envelope, according to observation by using negative stain after isolation and purification, was 20nm in length. The virogenic matrix was inside the cytoplasmic vesicles with membrane. Virion development in the cytoplasmic vesicles could be divided into three stage, viz, emergence, assembly and release. Infected cell with virion could bring about obvious pathologic changes, such as chromatin marginated and perinuclear space dilated in hypertrophied nuclei, ribosome detach from turgescent rough endoplasmic reticulum, myelinbody appeared in the cytoplasm, mitochondria swell and mitochondrial cristae rupture. Lastly, cellular disintegration resulted in empty space amongst the lesions. The pathological observation verified that the pathological effects were associated with the viral infection." @default.
