- W1018797730 abstract "Publisher Summary This chapter describes the derivation of monoclonal antibodies against the transferrin receptor and assay of their biological effects. Antibodies that block receptor function have proved to be useful in dissecting the role of transferrin receptors and iron uptake in cell growth. Monoclonal antibodies against the transferrin receptor of human, murine, and raw cells have been obtained. With one exception, all these antibodies were derived from fusions of spleen cells from mice or rats immunized with viable whole xenogeneic cells, usually cultured hematopoietic cell lines or activated lymphocytes. For most cell types, transferrin is the major source of iron, and transferrin-mediated iron uptake takes place via receptor-mediated endocytosis. After release of iron under acid conditions within the endosomal membrane compartment, apotransferrin remains bound to the transferrin receptor and is recycled back to the cell surface. The expression of transferrin receptors on growing cells and their obligatory requirement for transferrin suggests that this iron transport system and iron itself are important for cell growth." @default.
