- W1019205909 abstract "Publisher Summary This chapter discusses the coding behavior in the Escherichia coli in vitro protein-synthesizing system of fractions of bacterial RNA isolated by sucrose gradient centrifugation or by chromatography on methylated Albumin–Kieselguhr columns. The class of bacterial polynucleotides is likely to contain not only messenger RNA (mRNA) proper but also ribosomal RNA precursor molecules. Although no direct analysis has been made so far of the programming of protein synthesis in vitro by rapidly renewable ribosomal RNA precursors, it has been suggested that such RNA may direct the synthesis of a specific class of proteins—namely, ribosomal proteins. The protein synthetic stimulating capacities of bacterial RNA preparations may in part reflect the activities of components other than mRNA. The sedimentation behavior of the polynucleotides endowed with the capacity to stimulate protein synthesis in subcellular systems has been examined, using the technique of centrifugation on linear sucrose gradients. These have all revealed parallel distributions of biological activity and rapidly labeled RNA. The chapter discusses the preparation of unlabeled and rapidly labeled bacterial RNA, fractionation of rapidly labeled bacterial RNA, and biological activity of RNA fractions." @default.
