- W1029378448 abstract "Objective To study the expression of caspases-3, NF-κB and TNF-α in retina of diabetic rats and investigate the effect of EPO on these cytokins expression and their possible roles in diabetic pathogenesis. Methods Diabetes was induced on 56 Wistar rats by intraperitoneal injection of streptozotocin (STZ) at a dose of 60mg/kg.Diabetes model was characterized by an increase of rat blood glucose (>16.65mmol/L) in 24-48 hours after injection. The established diabetic rat models were randomly divided into diabetic mellitus (DM, 24 rat models) group and erythropoietin (EPO. 24 rat models) group, and 5000 U/kg of EPO was intraperitoneally injected in the latter group. According to the duration of diabetes, DM group and EPO group were subdivided into DM1, DM3, DM6 and EPO1, EPO3, EPO6 groups respectively. 0.9% sodium chloride was intraperituneally injected in DM group and 8 normal Wistar rats (negative control). The expression of caspases-3, NF-κB and TNF-α was studied by immunohistochemistry. Results No caspase-3 positive cell was observed in noriual retina and DM1 group. In DM3 group, positive expression of caspase-3 was found in ganglion cell layer (GCL). And in DM6 group, positive expression extended to the inner nuclear layer (INL). The positive expression of NE-κB was faint and only located in INL and ONL of normal retina, and similar findings were seen in DM1 rats. The expression of NF-κB was gradually increased in DM3 and DM6 rats, showing that the positive reaction mainly was in INL, ONL and GCL in DM3 rats and extended to almost the whole retina in DM6 rats. Under the intervention of EPO, the expressions of caspase-3, NF-κB and TNF-α were inhibited in various EPO groups. The distinction of positive expression between DM3 and EPO3 or DM6 and EPO6 base statistically significance (P<0.05). Conclusion The expression of caspase-3, NF-κB and TNF-α indiabetes was elevated which may take part in the early events of the onset of diabetic retinopathy. The Conclusion above may provide a theoretical basis fur the management of diabetic retinopathy using EPO." @default.
