- W102971932 abstract "Glycobiology is now ascending to its rightful position as a major research area for the life sciences. To keep pace with the increasing demands for characterization of biomolecules and for the elucidation of structure-function relationships among carbohydrate-containing species and their interactive partners, high-sensitivity mass spectral methods must be developed — broad enough to encompass distributions of related molecules that occur as natural mixtures, and yet specific enough to elucidate the details of composition, order and substitution patterns of each member in the series, some of which may be isomeric. We are using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) as our primary tool for the determination of molecular weights of glycoconjugates at the pmol level and below, and combinations of chemical derivatization, chemical and enzymatic degradation and tandem mass spectrometry to probe individual structures. Matrix-assisted laser desorption/ionization is quite tolerant of the presence of buffers and salts, and can be used to follow the course of reactions while consuming only low fmol amounts of sample. Under conditions of threshold irradiation, it results in little fragmentation and facilitates molecular weight determinations even when the intact species are too unstable for LSIMS detection. At higher irradiance, structurally significant fragmentation can be induced." @default.
