- W1034172024 abstract "This chapter presents the methods needed for characterizing nucleic acids encoding Aplysia calmodulin. It describes methods for dissection of several Aplysia tissues for isolating nucleic acids. The preparations obtained are suitable for analysis by hybridization techniques, and Northern, RNA dot-blot and Southern blot analyses have been used to examine the structure of the calmodulin gene and its expression in Aplysia. Genomic Southern blot analysis reveals only a single gene. Examination of calmodulin mRNA by Northern blot analysis indicates that mRNAs of 2.5, 3.0, and 3.8 kbp are transcribed from this single gene in nervous tissue. Aplysia differs from the other animals examined because the 3.8kb mRNA is specifically expressed in nerve cells of Aplysia, and is not detectable in muscle and other body tissues. The methods described for examining the gene structure and expression of calmodulin are useful for studying the gene structure and expression of other important Aplysia nerve cell proteins for which nucleic acid probes are becoming available." @default.
