- W1036195570 abstract "This chapter describes strategies for obtaining mutations in genes of selected membrane transport proteins. The emphasis is on strategy, not tactics. The chapter emphasizes on mutations that completely prevent production of a specified protein, or that profoundly alter its relative molecular weight, so that the absence of the protein can be recognized after separation of the membrane proteins by gel electrophoresis. Sensitive methods for recognition are addressed in the chapter. The examples discussed in chapter are restricted to sugar transport proteins of Escherichia coli. In all experiments involving sugars it is important to remember that even the best commercial preparations are not absolutely pure. Disaccharides are readily hydrolyzed in solution. Sugars are difficult to recrystallize without substantial losses. However, in principle the methodologies are applicable to any membrane protein (or even cytoplasmic protein) in any organism in which a combination of genetical and biochemical approaches is feasible." @default.
