- W1036257375 abstract "This chapter describes a general approach for the design of specific, potent reversible enzyme inhibitors. The inhibitors that are prepared with this approach are called transition state analogs. A transition state analog for an enzyme is a compound that resembles in structure the substrate portion of the transition state of the enzymatic reaction. Adenosine deaminase catalyzes the hydrolysis of adenosine to inosine, probably through a tetrahedral-like transition state formed by the attack of water. 1,6-Dihydro-6-hydroxy-methylpurine resembles the tetrahedral-like transition state by virtue of the H on N5, the tetrahedral geometry of C6, and, in one conformation the position of hydroxyl group. A possible transition state analog for chymotrypsin is 2-phenylethaneboronic acid, which is an effective competitive inhibitor of the enzyme. There is a similarity between the structure of an enolate anion and a carboxylate group, and several enzymatic reactions in which the transition state probably resembles an enolate anion are strongly inhibited by the analogous carboxylate anion. The enzyme that catalyzes the transfer of a phosphoryl group from adenosine triphosphate to creatine forms a very stable quarternary complex with adenosine diphosphate (ADP), creatine, and nitrate." @default.
