- W1038197168 abstract "This chapter discusses the formation of pneumococcal type XIV polysaccharide. Type XIV pneumocoecal polysaccharide (SXIV) is a highly branched polymer, composed of N-acetyl-D-glucosamine, D-glucose, and D-galactose in ratios of 2:1:3. Immunochemical studies suggest that the principal antigenic determinants of the polysaccharide are galactosyl end groups. A particulate enzyme preparation obtained from Type XIV pneumococcus catalyzes the synthesis of antibody reactive polysaccharides from the uridine diphosphate (UDP) derivatives of the sugars. Although the complete incubation mixtures yield polymers that resemble SXIV isolated from whole cells, careful immunochemical studies show that the enzymatic products are different. In addition to the synthesis of polysaccharides, the particulate preparation catalyzes a rapid synthesis of two glycosyl diglycerides; one of the assays for the polysaccharide synthetase system involves removal of the glycolipids. The particulate preparation is incubated with the sugar nucleotides, one of which is labeled, and polysaccharide synthesis followed by measuring the rate of incorporation of the labeled sugar into the polymer.
