- W1038225442 abstract "Publisher Summary This chapter discusses the sensitive measurements of changes of hydrogen ion concentration. Biological reactions involved in the production or consumption of acid or base can be studied easily by measuring pH. In the measurement of pH changes in biological reaction system, the amount of H+ produced or consumed, and the actual pH change is often necessary. Adequate stirring is necessary for the uniform mixing of components and for the rapid response of electrodes. Stirring of the reaction mixture is usually done mechanically, that is, by a vibrating rod, a magnetically driven flea, or a slow rotation of the reaction cuvette by a micromotor. Besides the common causes of errors in pH determination , the following are often encountered as possible causes of errors in studies of pH change with a high sensitivity: (1) dissolution of volatile acid, especially carbon dioxide, (2) electrostatic disturbance by charged objects, and (3) illumination of electrode. Many meters can be satisfactory for recording pH changes if this is the only quantity being recorded. However, more complicated devices currently in use where oxygen, potassium, and sodium and hydrogen ions are measured lead to interaction between the electrode, which is best controlled by allowing each electrode system to float, isolated from ground, and not connected to ground even at the recording end. The spectrophotometric method for determination of small changes of hydrogen ion concentration is admirably adapted to use with flow apparatuses, and here shows the great advantages over the glass electrode method, particularly with respect to response speed." @default.
