- W1038406670 abstract "This chapter discusses the development and modification of several double immunostaining techniques that can be combined with other methods, including degeneration and antero- and retrograde tracer techniques, to elucidate a substantial part of the anatomical basis of gonadal steroid-induced signal transmission to luteinizing hormone-releasing hormone (LHRH) neurons. Tissue preparation for electron microscopic (EM) double immunostaining reflects a compromise between the mutually exclusive requirements of structure preservation and deep penetration of antisera or antibodies. Although there are no specific studies analyzing the effects of different anesthetics on the outcome of single or double immunostaining, it seems that all of the most commonly used drugs, including ether, methoxyflurane, ketamine, pentobarbital, and chloropent, can be applied with the same satisfactory results. Detergents such as Triton X-100, saponin, or protease have been used to permeabilize sections, allowing for greater penetration of immunoreagents. Numerous EM double immunostaining techniques have been developed using two contrasting electron-dense labels. Under certain conditions, when two tissue antigens are localized in different, well-defined compartments of a cell, the same immunolabel can be used to visualize both substances." @default.
