- W1042370910 abstract "This chapter presents the studies of plant membranes by transmission electron microscopy (TEM). TEM study of cell membranes is a usual application for plant cell description. When working on ultrathin sections of resin-embedded biological material, with TEM, the limiting factor in obtaining ultimate resolution is by no means the electron microscope itself, but depends upon previous processing of the specimens. The point-to-point resolution of most TEM (often less than 0.2 nm) is far better than necessary for the optimal resolution of plant cell membranes. However, it must be remembered that high-resolution observations on membranes require an optimization of the electron microscope adjustments—that is, correct alignment of the lens system, choice and cleanliness of condenser and objective apertures, stability of the electron beam, presence of an effective anti contamination device, correction of lens astigmatism (condensers, objective), and optimal photographic records." @default.
