- W1044186016 abstract "This chapter discusses the procedure used to isolate P700-chlorophyll a -protein fractions from several species of higher plants, algae, and cyanobacteria. The P700-chlorophyll a -protein comprises the heart of photosystem I. It is believed to be ubiquitous in all chlorophyll a -containing plants with the possible exception of some photosystem I-deficient mutants. Depending on the plant source, 10–18% of the chlorophyll in higher plant chloroplasts is found in this pigment–protein. In the isolation procedure, chloroplasts are isolated from young leaves by differential centrifugation. The chloroplasts are suspended in a hypotonic buffer consisting of 0.1 M NaCl, 50 m M Tris-Cl, 20 m M sodium ascorbate, and 1 m M EDTA (pH 8.0) with the aid of a glass homogenizer. The lamellae are pelleted by centrifugation at 40,000 g for l0 min, resuspended in the same buffer, and again pelleted. These washed lamellae can be used immediately or suspended in a minimal volume of 50 m M Tris-Cl buffer containing 20 m M sodium ascorbate and 5 m M MgCl 2 (pH 8.0) and stored below 0°. The recovery of P700-chlorophyll a -protein from a typical experiment is shown. The yield of P700 and chlorophyll in the 0.2 M sodium phosphate eluate varies between 30–55% and 3–5%, respectively.
