- W1044411018 abstract "Publisher Summary This chapter describes the purification of alpha toxin from clostridium perfringens. Alpha toxin was the first bacterial toxin to be identified as an enzyme that hydrolyzes lecithin to phosphorylcholine and 1, 2-diglyceride. The toxin has lethal, necrotizing, and cytolytic activities and is considered to play the most important role in the pathogenesis of gas gangrene. Clostridium perfringens phospholipase C is a potential tool for studying membrane structure and function because of its high specificity for phosphatidylcholine as compared to sphingomyelin. This enzyme was found to be a zinc metalloenzyme. Calcium plays an essential role in the interaction of phospholipase with the substrate and provides the appropriate charge density. Clostridium perfringens produces, in parallel with phospholipase C, large amounts of toxins and enzymes that contaminate most preparations of alpha toxin. The two methods described in the chapter permit the preparation of Clostridium perfringens phospholipase C that is free of other toxins, particularly theta toxin (perfringolysin O) and delta toxin (in type C strains), which are both hemolytic and cytolytic." @default.
