Talab1 1 Urmia University, Department of Biology, Faculty of Sciences, Iran 2 Urmia University of Medical Science, Department of Physiology, Iran Electrical evoked responses of Schaffer collaterals of CA3 region of hippocampus is used for long time for different researches such as Long Term Potentiation (LTP). It seems that some drugs can affect LTP by changing component characters.Bupropion is an antidepressant drug that is being used to help in giving up smoking. In this study Electrically evoked population spikes(PS) were tested under the influence of perinatal bupropion(40mg/kg) exposure from stratum pyramidal of area CA1 of the hippocampus.300-400 μm thick hippocampal slices were taken from 19-25 days aged Wistar rats and maintained in holding ACSF for 60min. The slices were perfused by normal ACSF (300 mosmol, pH=7.4 in 95% O2 and 5% CO2 aeration) initially and after reaching to stabled response, Test stimuli were generated by isolated pulse Stimulators model with an insulated bipolar tungsten electrode Placed in the Schaffer collateral pathway.the Input-Output (I/O) curve was estimated to evaluation of 50% of maximum synaptic response.The responses acquisitioned and then the ACSF that contain Bupropion 10,50,200 Micromol was used for 30 min for treatment. The evoked responses were collected in the end of Bupropion-ACSF perfusion and then perfusion was changed to normal ACSF again. The results showed that Concentrations of 50,200 Mm perfusion bupropion reduced PS amplitudes in all slices (n=26) and after normal ACSF perfusion it took 1.5h ±20 min that responses to get back to initial status. we found adaptation synaptic in perfusion200 ìm compare with bupropion trated slices and non- treated. The results showed that there are some mechanisms involve in production of PS can affect by Bupropion. It seems that perkaryal synapses or interneurons can made same responses.
