- W106208510 abstract "For the past twenty-five years, the predominant neurochemical hypothesis associated with schizophrenia has involved a putative disturbance of dopamine neurotransmission. This model has undergone modification and refinement over the years, but the dopamine receptors have consistently been implicated in this disorder (Creese et al. 1976; Seeman et al. 1975; Snyder 1976). Classically, dopamine receptors were resolved into two subtypes, D1 and D2, based on pharmacological, behavioral, and neuro anatomical data (Attie et al. 1980; Stoof and Kebabian 1984; Memo et al. 1986; Onali et al. 1985). Most traditional antipsychotic drugs have a high affinity for the D2 subtype of dopamine receptor, and there is a well-established relationship between typical daily dose of these drugs and affinity for the D2 receptor (Creese et al. 1976; Seeman et al. 1975; Snyder 1976). These observations have been interpreted to suggest a primary alteration in the D2 receptor in schizophrenia or, at the very least, that the D2 receptor is a critical site of action for the antipsychotics." @default.
