- W107521208 abstract "Cardiac hypertrophy is an adaptive process characterised by cellular and metabolic remodeling. A major feature of this remodeling process is a reduction in fatty acid oxidation. Although initially beneficial, this may cause an inappropriate accumulation of intramyocardial lipids resulting in contractile dysfunction and heart failure. We investigated whether a mismatch between fatty acid supply and utilization occurs in cardiac hypertrophy that may predispose the heart to functional decline. Pressure overload hypertrophy was surgically induced in male Sprague-Dawley rats by abdominal aortic constriction (HT) over a period of 9 weeks. Group 1 received normal diet throughout, while Group 2 was fed high-fat diet (45% kcal from lipids) for the last 7 days. Hearts were isolated and perfused in an isovolumic Langendorff mode using a mixture of substrates in physiological concentrations; 5mM 113C glucose, 1 mM 313C lactate and 0.3mM U13C palmitate in 3% albumin. Cardiac hypertrophy was observed in HT compared to controls [Heart weight/Body weight ratios (g/Kg) - Group 1, 5.3 ± 0.5* vs 4.6 ± 0.4; Group 2, 4.9 ± 0.6* vs 4.2 ± 0.6, * p < 0.05]. High-fat diet increased fatty acid utilisation in HT hearts with a concomitant reduction in cardiac efficiency [(11.2 ± 2.9* vs 14.8 ± 4.7) X 103mm Hg /μmoles/ gwet weight, * p< 0.05 compared to HT hearts on normal diet].
