- W109598509 abstract "The strain Methylobacterium rhodesianum Z belongs to the group of “pink-pigmented facultative methylotrophs”. The bacteria are able to accumulate up to 45% poly-β-hydroxybutyric acid (PHB) under nitrogen-starved conditions with methanol as the sole carbon and energy source. By nitrogen-limited cultivation of the cells in pH-stat a growth-associated accumulation of PHB is attained. The amount of polymer increases from 2 % to 35 %. At the beginning of this phase the rate of product formation is very high. Following nitrogen starvation causes a second maximum of the rate of PHB synthesis. By combination of nitrogen limitation and starvation a higher PHB content in comparison to the polymer formation under nitrogen deficiency is reached. Der zur Gruppe der “pink-pigmented facultative methylotrophs” gehörende Bakterienstamm Methylobacterium rhodesianum Z speichert unter Stickstoff- und Schwefelmangel wachstumsentkoppelt Poly-β-hydroxybuttersäure (PHB). Auf Methanol als einziger Kohlenstoff- und Energiequelle akkumuliert die Kultur bei Stickstoffmangel-Bedingungen bis zu 45 % PHB in der Zellsubstanz. Durch Kultivierung der Mikroorganismen im pH-Stat unter Stickstoff-Limitation gelingt die wachstumsassoziierte Speicherung dieses Reservestoffes mit hoher Produktbildungsrate. Der Polymergehalt steigt bis zu 35% an. Nachfolgender Stickstoff-Mangel bewirkt ein zweites Maximum in der Produktbildungsgeschwindigkeit und eine höhere Polymerkonzentration als nach alleinigem Stickstoff-Mangel." @default.
