- W110336429 abstract "Publisher Summary The capacity to release hydrogen peroxide is a useful biochemical correlate of macrophage activation. Hydrogen peroxide or closely related substances have been directly implicated as both microbicidal and tumoricidal factors in mononuclear phagocyte effector function. There are two requirements to detect substantial hydrogen peroxide release from mononuclear phagocytes using the scopoletin method. First, if mouse peritoneal macrophages are employed, they must be activated. However, blood monocytes can secrete copious H 2 O 2 without prior exposure to activating stimuli. Second, the mononuclear phagocytes must be exposed to a triggering agent, that is, any variety of membrane-active stimuli, such as tumor promoters, antigen–antibody complexes, or phagocytic particles, which elicit a rapid response. Phorbol myristate acetate (PMA) is a triggering agent. PMA is an extremely potent tumor promoter. It must not come into contact with skin. Gloves should be worn throughout the assay and mouth pipetting should be strictly forbidden. Stock solutions should be tightly capped and thawed as briefly as possible. PMA-containing solutions should be disposed of according to institutional guidelines for hazardous chemicals. The PMA waste bottle in the laboratory should be nonbreakable and should not be used for the concomitant storage of carcinogens." @default.
