• W112263027 abstract "We have comparatively investigated some biochemical and pharmacological properties related to recognition of DNA AT-rich sequences of two hybrid groove binder-intercalators netropsin-oxazolopyridocarbazole (Net-OPC) and bis-pyrrolecarboxamide-oxazolopyridocarbazole (BPC-OPC) in which the amidine moiety of netropsin has been replaced by a methoxy group (Mrani et al. 1990 submitted). The hybrid molecule Net-OPC binds to poly d(A-T) at two different sites with Kapp values close to 7 106 and 6 108 M-1 (100 mM NaCl, pH 7.0) whereas it binds to poly d(G-C) to a single binding site with a lower Kapp of 1.7 106 M-1. In contrast, the molecule devoid of the amidine moiety BPC-OPC binds to both poly d(A-T) and poly d(G-C) with quite similar Kapp values of 2.2 106 and 1.4 106 M-1 indicating that the presence of the amidine is required for recognition of AT-rich sequences. Based on viscometric data, the binding of Net-OPC is suggested to involve both intercalation and external binding of the OPC chromophore in AT-rich sequences and intercalation only in G-C-rich sequences. In similar experimental conditions, BPC-OPC binds to AT and GC-rich sequences according to an external mode." @default.
