- W1132488462 abstract "Publisher Summary This chapter discusses the in vivo labeling of retinal components by intraocular injection of the isotopes. In this technique, the quantity of isotope needed is reduced by a factor of about 1000. The savings can be highly significant when the procedure is carried on larger animals, such as cats, dogs, or primates. An additional advantage of intraocular injection is the ability to use each eye independently because there is little crossover of radiolabeled precursor from one eye to the other. In an attempt to follow intracellular transport of macromolecules or metabolic processing of precursor molecules, it is important to quickly label the pool of molecules and to ensure that no further labeling should take place—that is, the pool should be pulse labeled. This permits uncomplicated analysis of further migration or metabolism of the pool. Intraocular injection accomplishes pulse labeling of retinal components when viewed on the scale of a few hours. The most convenient technique for rapidly anesthetizing a series of rats or mice for sequential injections involves the use of an inhalation chamber. For the injection of radiolabeled compounds, a 10-μL Hamilton (Reno, NV) syringe is used." @default.
