The Xenopus oocyte system is perhaps uniquely suited to the study of ion channels and neural receptors in a controlled in vivo environment. The oocyte system is used as an assay for the isolation of complementary DNA (cDNA) clones encoding receptors for which the only information available was a functional response. The oocyte expression system is particularly useful for the isolation of cDNA clones encoding ion channels for which no sequence information is available. Two basic approaches are used: (1) hybrid selection or depletion, and (2) direct expression. There are a number of important considerations in using Xenopus oocytes to study ion channel and receptor functions. The first of these is that uninjected oocytes express a variety of electrophysiological responses which might interfere with the one being studied.
