- W1161591634 abstract "Publisher Summary This chapter summarizes the types of experiments employing reconstituted vesicles in the study of the H+ pumping activity of cytochrome oxidase. The most extensive measurements of H+ translocation by cytochrome oxidase vesicles are those where a small number of enzyme turnovers is induced, either by the addition of reduced cytochrome c to an aerobic sample, or of O2 to an anaerobic sample in the presence of excess reduced substrate. A typical reductant-pulse measurement using phenol red as the H+ activity monitoring system is described. A crucial test of H+ translocation by cytochrome oxidase that is not affected by any hypothetical proton-release artifacts is the determination of the charge-translocation stoichiometry. Measurement of charge translocation may be achieved using experimental approaches such as reductant pulse, O2 pulse, or rate measurements. In addition, an ion-sensitive electrode is employed to monitor the uptake of extra-vesicular permeant cations, such as K+ (in the presence of valinomycin) or tetramethyl-p-phenylenediamine (TPP+)." @default.
