Enzymatic reaction is a physiologically relevant biochemical stimulus for the temporal and spatial control of drug release because: (1) the overexpression of specific enzymes is localized at diseased tissues; (2) the activity of disease-associated enzymes is correlated with the progression of the disease; and (3) enzymes exhibit specific substrate preference. Enzyme-sensitive biomaterials are designed for constructing targeted drug-delivery systems, with examples in polymer–drug conjugates, hydrogels, liposomes, polymeric particles, and self-assembling nano- and macrostructures. The common theme in material design involves the incorporation of specific enzymatic substrates as structural components. This allows a change of material property upon an enzymatic reaction that leads to enzyme-mediated drug release. This article details the strategies and technical issues for synthesizing enzymatic triggers in various drug-delivery systems, including those using enzyme substrates as pendant groups, cross-linkers, and integral parts of self-assembling building blocks." @default.
