- W11721758 abstract "This chapter discusses a study on the cytotoxic activity of the monocytes and its potentiation by lymphokines and interferon. In a study described in the chapter, cytolytic activity of the monocytes was measured by 51Cr-release from prelabeled tumor target cells after 3–4 h of interaction with effector cells. The cytolytic activity was studied at the single-cell level with the conjugate/agarose technique using a modification of the method described for cytotoxic T lymphocytes. Monocytes and tumor target cells at 1:1 ratio were incubated in a pellet for 10 min at 30°C and 20 min at 4°C. The cells were then gently resuspended in molten 0.5% agarose at 39°C and quickly spread on a microscope slide. After incubation for 3–4 h at 37°C, the slides were incubated with trypan blue to stain the dead cells. Each slide was scored for the percent bound monocytes and the percent dead conjugated target cells, and the number of cytotoxic monocytes was calculated. It was found that the various human suspension cell lines used as targets differed in their sensitivity to lysis, with K562, a myeloid cell line, being the most sensitive one.
