The realization that polyisoprenoids were synthesized from a C 5 precursor, isopentenyl pyrophosphate, prompted speculation as to the mechanism of the condensation reactions involved. The isomerase may also be involved in the synthesis of nonpolyisoprenoids. Dimethylallyl pyrophosphate may be the immediate precursor of the dimethylallyl group found attached to nitrogen or carbon in various natural products. This suggestion has been supported by the incorporation of radioactivity from radioactive dimethylallyl pyrophosphate into N 6 -dimethylallyladenosine in tRNA by a partially purified enzyme from yeast. All the assays of isopentenylpyrophosphate isomerase depend upon the availability of radioactive isopentenyl pyrophosphate. The substrate, isopentenyl pyrophosphate, is stable to mild acid treatment. The radioactive alcohols can be isolated by extraction with ether or light petroleum and the extract assayed for radioactivity. The amount of isomerization can be deduced by estimating the amount of radioactive isopentenyl pyrophosphate remaining after the acidification and removal of the dimethylallyl alcohol and dimethylvinyl carbinol." @default.
