- W1186643035 abstract "This chapter describes the preparation, storage, use, and properties of dynein ATPase from sea urchin sperm flagella, which is probably one of the best available sources for providing relatively large quantities of this enzyme. Selective extraction and recombination followed by electron microscopy indicate that dynein comprises part or all of the arms bridging the gap among the doublet microtubules of the axoneme. High molecular weight, possession of ATPase activity, extractability with low-ionic-strength solutions in the presence of EDTA, and relationship to the arms on doublet tubules have become the identifying characteristics of dynein from other sources. The methods described in the chapter relate specifically in preparing latent activity dynein-1 (LAD-1) from sperm flagella of the Hawaiian sea urchin Tripneustes gratilla. The general techniques are applicable to sperm flagella of many other animals. A brief comparative review of the methods that have been used for the preparation of dynein from various other sources and a list of some of the salient properties of these dynein ATPases are also provided in the chapter." @default.
