- W12034750 abstract "Suspensions of human embryonic stem cell-derived retinal pigment epithelium (hES-RPE) and human fetal RPE (fRPE) resurface aged and age-related macular degeneration (AMD) Bruch’s membrane (BM) to a limited degree at day 21 in organ culture. The presence of AMD-related changes, such as basal linear deposit, is associated with worse BM resurfacing. Survival and differentiation of hES-RPE and fRPE on aged or AMD BM is enhanced greatly (200 %) if a biologically synthesized extracellular matrix (ECM) (bovine corneal endothelial cell (BCEC) ECM) is laid down on BM prior to transplantation. Transplanted RPE survival is enhanced even more (400–1,000 %) if BM is treated with BCEC conditioned medium during organ culture of hES-RPE or fRPE on aged or AMD BM. Cells so treated also produce a variety of neurotrophic factors (e.g., brain-derived neurotrophic factor, pigment epithelium-derived factor, nerve growth factor) while on BM even if they do not exhibit highly differentiated morphology. This neurotrophic factor production may contribute to improved vision that has been observed in some patients who have received allogeneic hES-RPE transplants. Future efforts are focused on identifying the bioactive components of BCEC-CM so that this material can be reconstituted for clinical use as an adjunct to improve RPE transplant survival and differentiation in AMD eyes." @default.
