- W1239628081 abstract "This chapter discusses the preparation and purification of formiminoglycine formimino transferase. 5-Formiminotetrahydrofolic acid formed in the enzymatic reaction is converted to 5,10-ethenyltetrahydrofolic acid in the presence of an excess of formiminotetrahydrofolic acid cyclodeaminase. The 5,10-methenyltetrahydrofolic acid formed is determined by its absorption at 356 mμ, and the rate of its formation is a function of the concentration of formiminoglycine formimino transferase. The presence of cyclohydrolase interferes with the present assay, as it hydrolyzes the product to 10-formyltetrahydrofolic acid, which has decreased absorbance at 356 mμ. The amount of interference encountered in natural products, however, will depend on the relative amounts of the formimino transferase and the cyclohydrolase. In cases investigated, the cyclohydrolase is relatively inactive and does not seriously interfere with the assay of formimino transferase by this method. The enzyme is entirely specific for formiminoglycine. The compounds tested and found to be completely inactive are formiminoalanine, formiminoglycinamide, formiminoglycine methyl ester, formiminoglutamic acid, formiminoaspartic acid, and formylglycine." @default.
