• W1267988237 abstract "Publisher Summary A large number of studies have been reported that exogenously concluded that gangliosides can have positive effects on neurite outgrowth. The molecular mechanism underlying these positive effects, however, remains unclear and it has often been difficult to determine whether gangliosides mimic the full range of trophic effects of neurotrophic factors such as nerve growth factor (NGF) or whether they have more limited effects. In dorsal root ganglion neurons—that are NGF responsive—GM1 could enhance some NGF effects but that it could not substitute for NGF by directly stimulating neuronal survival. It therefore seems likely that gangliosides act by modulating signals that lead to neurite outgrowth and do not by themselves possess full neurotrophic activity. Moreover, it is equally likely that GMl modulates integrin or cell adhesion molecules (CAM) signals rather than NGF signals per se . To analyze the possible interactions of gangliosides with CAM signaling pathways, the chapter presents a series of co-cultures of PC12 cells with monolayers of 3T3 fibroblasts or the same monolayer cells expressing transfected NCAM or N-cadherin. It is shown that a direct incorporation of GM1 into the cell membrane of PC12 cells—in a trypsin insensitive manner—is associated with a dramatic enhancement of NCAM and N-cadherin-dependent neurite outgrowth." @default.
