It deals with the morphogenesis of the myoneural junction of rat, but for comparison, the corresponding human myoneural morphogenesis is also discussed. The structural development of the cholinergic myoneural synapse is a relatively complicated phenomenon that can be studied by histochemical methods based on the demonstration of functionally important components. A histochemically demonstrable acetylcholinesterase (AChE) activity of the myoneural junction is localized in the postsynaptic membrane, revealing the structure of this membrane during the development. The motor axon first ramifies into many terminals that are apposed to a smooth muscle membrane. Subsequently, a depression is formed in which the postsynaptic infoldings appear. The axon terminals are gradually separated from each other and a folded postsynaptic membrane surrounds each terminal, thus forming the structurally complex mature myoneural junction. Morphogenesis of the cholinergic synapse in striated muscle appears to be, in the main, a similar phenomenon in different species; only the intervals between the different stages of development vary. Based on experimental studies, it looks as if developing or regenerating nerves control the morphogensis of the cholinergic synapse in the striated muscle by a mechanism not involved in the neurotransmission. This morphogenesis of the myoneural junction provides a suitable model for experimental studies on the trophic functions of the neurone." @default.
