- W131535545 abstract "This chapter discusses a specific oviduct target-tissue receptor for progesterone. It presents the identification, characterization/partial purification, intercompartmental transfer kinetics and specific interaction with the genome. There are three lines of evidence that clearly suggest a role for the oviduct binding protein(s) in progesterone action. First, these molecules are tissue specific, having been found only in the oviduct and not in spleen, lung, intestine, liver or blood. In addition, these components selectively interact with progesterone-like compounds. The binding specificity has been determined using sucrose gradient analysis and also by a more convenient glass adsorption assay which is similar to that used by Clark and Gorski to measure estrogen binding in the rat uterus. In this assay, the hormone-labeled complex is separated from the unbound hormone by the ability of the complex to adsorb to glass particles at 23°C under conditions of high Mg++ (15 mM). Thus, the macromolecular-hormone complex can effectively be removed from the solution. The third line of evidence supporting a physiological role for the progesterone-binding molecules is based on the correlation of their concentration with the stages of induced oviduct development. Because the responsiveness of the oviduct to progesterone is enhanced by prior estrogen treatment, it was thought that the amounts of progesterone-binding protein could also be affected by estrogen. Indeed, the progesterone-binding capacity of the oviduct cytosol increases ten-fold during 17 days of the estrogen treatment." @default.
