- W135036770 abstract "The purpose was to determine the effects of PDK4 deficiency on the long-term consequences (hyperglycemia, glucose intolerance, hepatic steatosis, and obesity) of feeding mice a high-saturated fat diet. PDK4 deficiency might attenuate the negative effects of saturated fat by lowering blood glucose and improving insulin sensitivity, or it might promote fat accumulation in tissues and worsen insulin sensitivity by inhibiting fatty acid oxidation. This was examined by feeding mice a 60% saturated fat diet for 7 months. Hyperglycemia was induced in the wild type mice but attenuated in the PDK4 knockout mice. PDK4 knockout mice gained less weight. Glucose tolerance was improved and insulin sensitivity was greater in PDK4 knockout mice. Accumulation of fat in the liver of PDK4 knockout mice was reduced compared to wild type mice. PGC-1α was reduced while ACC and FAS were increased in the liver of wild type mice fed the saturated fat diet compared to wild type mice fed a chow diet. PGC-1α was not reduced and ACC and FAS were not increased in PDK4 knockout mice fed the saturated fat diet. These findings suggest that knocking out PDK4 creates conditions that prevent reduction of fatty acid oxidation and induction of fatty acid synthesis. This suggests that PDK4 deficiency exerts beneficial effects in saturated fat fed mice by altering upstream signaling components involved in regulation of fat metabolism.
