- W137305217 abstract "Undoubtly, cryptands remain the class of macrobicyclic and macropolycyclic receptors looking forward to a great future. The number of possible structures is practically unlimited and depends on a researcher imagination and creativity. Initially, the chemistry of cryptands had focused on the development of new synthetic strategies, either to improve the yields or to built up more complicated molecular architectures, like cylindrical, spherical or lateral macropolycycles, bi- or polynucleating, with hard and soft binding sites, etc. Simultaneously, extensive investigations concerning the physico-chemical properties of cryptands have been conducted covering several sub-topics: their complexing properties, X-ray analysis, molecular dynamics of inclusion complexes, etc. One important feature of cryptands is their enzyme analogy in certain instances, which could be very helpful to understand the natural enzyme activities. The next step in the chemistry of cryptands therefore will probably be the design, synthesis, and the study of the properties of more elaborated polyfunctionalyzed macropolycyclic structures allowing a deeper insight into the principles of enzyme reactivity. Actually in the synthetic respect, high-dilution methods enjoy a great popularity because of its generality. Nevertheless, as far as the introduction of sensitive groups into macropolycyclic architectures is concerned, the high-pressure method which offers milder reaction conditions than high-dilution techniques is among the most favoured alternatives, although the high-pressure method has one serious drawback, i.e. the availability of the apparative set-up.
