The use of fluxes rather than conventional pathways provides a very flexible framework that, together with the concepts of flux-generating and regulatory steps, provides a solid foundation for subsequent quantitative analysis. The use of intrinsic sensitivities, which are easily derived from kinetic and metabolic data, enables complex regulatory systems to be analyzed and computed. The computation is straightforward and is much simpler than attempting to solve a set of full rate equations: all detailed computations are relegated to adjusting the value of matrix elements at the end of each stage of the numerical procedure. However, computation using sensitivity functions does require knowledge of the initial conditions, and these must either be calculated using full rate equations or must be, preferably, obtained by experiment. Unfortunately, the experimental determination of fluxes and metabolite concentrations at the site of specific enzymes or enzyme systems is at present very difficult." @default.
