Methods described contain information regarding the excitation and emission properties of the dyes. However, as in designing new staining reactions or selecting combinations of multiple dyes for different cell constituents in a single staining mixture, there is a need for rapid review of the spectral properties of numerous probes. Most of the reagents that have application in flow cytometry (FCM) are derivatives of a smaller group of dyes, such as fluorescein, rhodamine, coumarin, acridine, xanthene, or thiazine. Many groups of reagents thus share similar spectral properties with these dyes. Chemical modifications changes spectral characteristics of the probe not only by shifting the excitation and/or emission band in either direction but also by altering the luminescence quantum yield. Caution should be exercised in projecting the spectral properties of the derivatives based on spectral data of the parent dye." @default.
